That is the face of a man who knows something and is trying hard to not say it.

actors aren’t told future story-lines. Jensen’s even said he didn’t know Dean would become a demon until he got the script a few weeks before they shot the episode

I think that’s more the face of someone who’s been playing a character for so long that he’s not sure what life would be like without Dean. At comic con someone asked him what was the hardest thing about playing the same character for 10 years and he said “Someday I’ll have to say good-bye to him”
